چکیده

Polysomes involved in ovalbumin synthesis were identified by the binding of 1251-anti-ovalbumin to hen oviduct polysomes. Techniques were developed for the isolation of undegraded hen oviduct polysomes and for the preparation of 1251-y-globulin free of ribonuclease activity. The distribution of 1251-anti-ovalbumin in the polysome profile is in accordance with the size of the polysomes that are expected to be synthesizing ovalbumin. Anti-ovalbumin does not react with polysomes extracted from tissues that do not synthesize ovalbumin, and antibodies directed against a protein that is not synthesized in the oviduct do not bind to hen oviduct polysomes. These criteria indicate that the reaction is specific for polysomes that synthesize ovalbumin. Polysomes that bind 1251-anti-ovalbumin can be precipitated by the addition of ovalbumin and an excess of specific antibody.

Isolation of hen oviduct ovalbumin and rat live albumin polysomes by indirect immunoprecipitation.

The polyribosomes synthesizing ovalbumin and albumin have been isolated from total oviduct polysomes and total rat liver polysomes, respectively. The isolation was performed using an indirect immunoprecipitation technique which is based on the specific reaction which occurs between antibody against a purified native protein and the nascent peptide chains on polysomes synthesizing that protein. ...

Studies on the attachment of carbohydrate to ovalbumin nascent chains in hen oviduct.

We have examined the question, is the carbohydrate moiety of ovalbumin attached to nascent chains or to the completed protein after its release from polyribosomes. Ovalbumin nascent chains have been isolated from hen oviduct by immunoprecipitation of ovalbumin-synthesizing polysomes and subsequent isolation of peptidyl-tRNA by DEAE-cellulose chromatography. Our results indicate that both glucos...

Identification and purification of collagen-synthesizing polysomes with anti-collagen antibodies.

Antibodies against embryonic chick bone collagen were prepared in rabbits and were purified by affinity and ion exchange chromatography until collagen-specific and RNase-free. 125I-anti-collagen antibodies were used to locate the collagen-synthesizing polysomes of 8-day chick embryo wings and legs on sucrose gradients by measuring the polysome associated radioactivity. The 125I-anti-collagen an...

Isolation of specific messenger RNA by adsorption of polysomes to matrix-bound antibody

A procedure is presented for the purification of specific mRNAs, which exploits the ability of antibodies prepared against a native protein to bind to the nascent polypeptide on the polysome. Rather than precipitating these soluble antibody-polysome complexes with anti-antibody, which can lead to nonspecific trapping of polysomes, we have linked the anti-antibody to an insoluble matrix. Thus, t...

Regulation of Protein Synthesis in Chick Oviduct

“Superinduction” is a phenomenon frequently observed in eukaryotic cells, whereby the inhibition of RNA synthesis (usually with actinomycin D) results in an increase in the concentration (activity) of a specific protein. This paper shows that actinomycin D can superinduce the major egg white proteins synthesized in chick magnum. After actinomycin D treatment, ovalbumin, conalbumin, ovomucoid, a...
